Genetic heterogeneity, genomics and risk prediction

Genetic variability influences the phenotypic manifestation of complex traits that are also modifiable by environmental effects. The increasing ability of laboratory scientists to detect and identify such variation must inevitably change the manner in which health professionals predict risk of partially hereditary disorders and in addition, how they design and interpret clinical trials. Apparently contradictory findings of clinical trials which are not resolved by meta-analysis might be attributable to genetic heterogeneity in the pool of participants. Our models attempt to identify effects of heterogeneity on key disease outcomes and also suggest alternative approaches to risk prediction and clinical trial design.
